Forget cooperativity! – single-molecule signalling with cGMP

چکیده

We have characterized two novel cyclic nucleotide-gated (CNG) ion channels that are distinctively different from classic vertebrate CNG channels. One channel is from the bacterium M. loti and the other from sperm of sea urchins. Two conspicuous features that set apart these channels from known CNG channels: [1] an exquisite ligand sensitivity and [2] the lack of cooperative activation. In my talk, I'll show that binding of a single cGMP molecule is sufficient to open these channels. The exquisite sensitivity of the sea urchin CNGK channel endows sperm with the ability to register a single molecule of chemoattractant during chemotaxis and to generate a brief Ca2+ signal. During evolution two different classes of CNG channels evolved. One class is designed to operate at "high" ligand concentrations (μM range); the other class is suitable to faithfully track ligand concentrations in the nanomolar range. At these low concentrations, the probability is miniscule that several cGMP or cAMP molecules simultaneously bind to the same channel. Therefore, a cooperative binding mechanism would impair sensitivity rather than enhance it.

Negative feedback in NO/cGMP signalling.

Most of the effects of the signalling molecule nitric oxide (NO) are mediated by the stimulation of the NO-sensitive GC (guanylate cyclase) and the subsequent increase in cGMP formation. The enzyme contains a prosthetic haem group, which mediates NO stimulation. In addition to the physiological activator NO, NO-sensitizers like the substance YC-1 sensitize the enzyme towards NO and may therefor...

Single-channel kinetics of the rat olfactory cyclic nucleotide-gated channel expressed in Xenopus oocytes.

Cyclic nucleotide-gated channels are nonselective cation channels activated by intracellular cAMP and/or cGMP. It is not known how the binding of agonists opens the channel, or how the presumed four binding sites, one on each subunit, interact to generate cooperativity. We expressed the rat olfactory cyclic nucleotide-gated channel alpha subunit in Xenopus oocytes and recorded the single-channe...

Calcium Modulation of Ligand Affinity in the Cyclic GMP–gated Ion Channels of Cone Photoreceptors

To investigate modulation of the activation of cGMP-gated ion channels in cone photoreceptors, we measured currents in membrane patches detached from the outer segments of single cones isolated from striped bass retina. The sensitivity of these channels to activation by cGMP depends on the history of exposure to divalent cations of the membrane's cytoplasmic surface. In patches maintained in 20...

Cooperativity in biological systems

Living organisms can sense and respond to external and internal stimuli. Response isdemonstrated in many forms including modulation of gene expression profiles, motility, secretion, cell death, etc. Nevertheless, all forms share a basic property: they depend on sensingsmall changes in the concentration of an effector molecule or subtle conformational changes ina protein and invoking the appropri...

cGMP-dependent protein kinase as a modifier of behaviour.

The importance of cGMP-dependent protein kinase (PKG) to the modulation of behavioural phenotypes has become increasingly clear in recent decades. The effects of PKG on behaviour have been studied in diverse taxa from perspectives as varied as ethology, evolution, genetics and neuropharmacology. The genetic variation of the Drosophila melanogaster gene, foraging (for), has provided a fertile mo...
